Peripheral neuropathy is the result of damages of the peripheral nervous system. These troubles can be caused either by a trauma or diseases as for example diabetes.

Models of CHEMOTHERAPY INDUCED NEUROPATHY
Cisplatine, vincristine and taxol are widely used as antimitotic drugs for treating various cancers. However, their employment is often limited by their neurotoxicity
( Quasthoff et Hartung, 2002 ) . Counteracting this side effect without affecting their anti-cancer activities is a promising developmental strategy.

Compound testing
These models are usually proposed to assess the neurotoxicity of chemotherapies or to study neuroprotectant activities but other treatments could also be considered. Please feel free to contact us to discuss the feasibility of your study.

Purified cultures of sensory neurons are injured by chemotherapeutic drugs (cisplatine 3 µg/ml, vincristine 7.5 ng/ml or taxol 0.5 µg/ml). The aim of this study is to investigate the putative protective effect of tested compounds on this damage. Neuronal survival is assessed by measuring acid phosphatase activity after exposure to chemotherapy and tested compound. -
Co-cultures from sensory neurons and Schwann cells are injured by chemotherapeutic drugs (cisplatine 0.5 µg/ml, vincristine 1 ng/ml or taxol 0.5 µg/ml). The aim of this study is to investigate the putative protective effect of tested compounds on this damage. Global cellular survival is assessed by measuring acid phosphatase activity after exposure to chemotherapy and tested compound.

Purified cultures of sensory neurons :
Cisplatin induced cell death in sensory neurons.
At each dose of cisplatin, NGF at 5 ng/ml is able to protect neurons against the ciplatin damage. -
Co-cultures of sensory neurons and Schwann cells :
Cisplatin induces cell death in a dose dependant manner.
At each dose tested, NGF at 5 ng/ml is able to protect neurons against ciplatin damage.

Taxol induced sensory neurons death.
NGF is able to protect sensory neurons against this injury. -

Taxol induces sensory neuron cell death.
NGF is able to protect sensory neurons against this injury.

Vincristine induces cell death in sensory neurons in a dose dependant manner.
