Resveratrol reverses inflammation-induced cognitive decline

Newsletter # 92

In vivo studies

Resveratrol, a polyphenol naturally present especially in red wines and grapes is among phytochemicals investigated to explain the French Paradox, i.e., a relatively low rate of cardiovascular disease in the french population, despite a high intake of saturated fat. There are compelling evidences showing the antioxidant, anti-inflammatory and phosphodiesterase inhibitory effects of resveratrol.
Through these mechanisms, plethora of health benefits have been proposed for resveratrol including cardioprotective, antithrombotic, anticarcinogenic, and neuroprotective properties

Some number of clinical investigations showed that resveratrol supplementation can bring beneficial effects to patients with mild cognitive impairment (MCI) or Alzheimer's disease (AD)
(reviewed in DOI: 10.1089/jmf.2021.0084).

  • ***, p ≤ 0.001 significantly different as compared
to LPS / Vehicle group (n = 10 mice per group)
  • At Neurofit, 1 week (once daily) treatment with resveratrol fully restores inflammation-based cognitive deficit in the mouse (see below).
This preclinical finding further supports the potential therapeutic benefits of resveratrol in the brain disease conditions involving inflammation such as AD.

The graph shows a marked reduction in the level of spontaneous alternation of LPS-mice (red column) during the T-maze task as compared to that of control mice (grey column). The decrease in the alternation reflects stereotypic and repetitive behavior, a common symptoms in dementia patients.
    (DOI: 10.1177/1533317513481094).

The treatment of LPS-mice with Resveratrol (green column) significantly increases the alternation performance of LPS-mice. Resveratrol was administrated at 10 mg/kg once daily for 7 days.
