Cellular model of brain inflammation and neuronal death

Newsletter # 42

Triple immunofluorescence images of neurons (nuclei stained in blue), microglia (green),and astrocytes (red) in LPS-activated cocultures.

Cellular models

Neuroinflammation is now recognized as a critical process in different neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, stroke, multiple sclerosis, etc... Astrocytes and microglia are key players in neuroinflammation since they release wide variety of proinflammatory mediators, including nitric oxide, cytokines and chemokines.

NEUROFIT has established a cellular model of brain inflammation where microglia-astrocyte-neuron triad is involved. This model is useful for screening the potential of compound to interfere with the neuroinflammatory process and to prevent neuronal death.

  • Release of neurotoxic factor such as nitric oxide (NO) in LPS-activated cocultures.

Resveratrol and dexamethasone inhibit in dose-dependent manner the NO release by LPS-activated cocultures.
  • Neuronal death in LPS-activated cocultures.

Resveratrol and dexamethasone dose-dependently inhibit neuronal death in LPS-activated cocultures.

NEUROFIT offers a range of validated in vitro and in vivo screening tests for psychiatry and neurology.
